How to use?
There are two methods for cell cryopreservation: the traditional method is to use cryopreservation tubes at 4 ℃, – 20 ℃ and – 80 ℃. The programmed cooling method is to use the programmed cooling box to set the program from room temperature to – 80 ℃, and then place it in the liquid nitrogen tank for long-term storage.
Experience of using program cooling box
1. When using, put the cell cryopreservation tube into the cryopreservation box stored at room temperature, and then place it in the – 80 ℃ cold storage environment, using isopropanol to achieve gradient cooling. Transfer to liquid nitrogen tank the next day, and transfer valuable cells as soon as possible, otherwise the survival rate of resuscitation will be affected. Generally, replace the isopropyl alcohol after 5 times of use.
2. The liquid level of isopropyl alcohol should be higher than the scale mark on the external surface of the freezing box, otherwise it can not play a very good role in program cooling.
3. The freezing box shall be stored at room temperature. If the freezing box is taken out from – 80 ℃, it shall be left at room temperature for a period of time until it recovers to room temperature before use.
4. Theoretically, isopropanol needs to be replaced after five times of use, but isopropanol is harmful to the environment, so we can use it several times (for example, 6-7 times). Isopropyl alcohol in the freezer is generally 250ml. If it is lower than the scale mark, add it. DMSO in the cryopreservation solution is toxic to cells, especially at room temperature, so the cryopreservation box and the cryopreservation solution should be put at 4 ℃ first, the cells should be resuspended with the cryopreservation solution, and then put in the cryopreservation box. The cells should be stored at 4 ℃, and then stored at – 80 ° when they leave the cell room. It will be transferred to liquid nitrogen tank the next day.
The gradient cooling function of the programmed cooling box can greatly improve the survival rate of cell cryopreservation. Due to the concentration of isopropanol and foam lining in the programmed cooling box, the programmed cooling box can also be used for cell resuscitation.
